Chain Adjustment

I am having trouble adjusting chain on wifes bike. Its K5 600 frame with 2003 1000 motor. Bike is slammed, with bolt on extensions, gearing is 16/45. Having pain in ass time trying to adjust chain out, got a whinning noise. When measuring, do I push down hard as I can, measure distance then push up hard as I can, measur distance then add the two together, giving me total slack? I am measuring with her setting on bike. What are ya'll doing, how are ya'll measuring, amount of slack? Chain seems tight, but there is total 1" can push down hard/up hard-get .5 total travel each way-adding up to 1".
Just got my extensions/gears on my ZX14, gunna race tomorrow-dont want mess something up-by adjusting it wrong.

Your reading out of the owners Manuel. That's for a swb chain. A extended chain is going to move more, because the two sprockets are further apart.
